Production and optimization of L-asparaginase in Escherichia coli

چکیده
-ASPARAGINASE (L-ASNase) has been widely used as a therapeutic agent in the treatment for various lymphoblastic leukemia diseases. This study aimed to isolate and purify local bacterial isolates that are capable of producing L-ASNase, so 150 bacterial isolates from the Nile River where isolated, purified and their ability to produce L-ASNase was assessed. Among these isolates, 32 bacterial isolates showed their ability to produce L-ASNase, so they were selected for further studies. The most active bacterial isolate in the production of L-ASNase was selected, identified as Escherichia coli and named MG27. Enzyme localization was determined in cultures grown under aerobic and anaerobic conditions. E. coli MG27 was found to produce more L-ASNase in anaerobic conditions compared with that produced under aerobic conditions with 128 folds. Finally, in an attempt to determine the optimum conditions for periplasmic L-ASNase production, the influence of several cultural factors was investigated.

Soluble Expression and Purification of Q59L Mutant L-asparaginase in the Presence of Chaperones in SHuffle™ T7 strain
Background and Aims: Q59L mutant of L-asparaginase enzyme from Escherichia coli (E. coli) has been introduced with lower side effects. This version of the enzyme might have potential applications in the treatment of leukemia patients. We utilized SHuffle T7 strain of E. coli, to produce the mutant enzyme in the presence of chaperone molecules. Materials and Methods: Q59LAsp gene was cloned in...
متن کاملProduction of L-Asparaginase by Serratia marcescens SB08: Optimization by Response Surface Methodology
This paper describes optimization method that combines the Plackett-Burman design, a factorial design and the response surface method, which were used to optimize the medium for the production of L-asparaginase by Serratia marcescens SB08. Four medium factors, from out of 11 medium factors, were screened by Plackett-Burman design experiments and subsequent optimization process to find out the o...
متن کاملNew procedures for purification of L-asparaginase with high yield from Escherichia coli.
l-Asparaginase is now known to be a potent antineoplastic agent in animals and has given complete remission in some human leukemias. Extensive clinical trials of this enzyme, however, were not possible in the past because of inadequate production of this substance. We have developed practical procedures for producing l-asparaginase in yields of sufficient quantity and purity for more extensive ...
متن کاملBioinformatic Analysis of L-Asparaginase II from Citrobacter Freundii 1101, Erwinia Chrysanthemi DSM 4610, E. coli BL21 and Klebsiella Pneumoniae ATCC 10031
Backgroung and Aims: L-Asparaginase II is a cornerstone of treatment protocols for acute lymphoblastic leukemia. Only asparaginase II obtained from E. coli K12 and Erwinia chrysanthemi have been used in human as therapeutic drug. The therapeutic effects of asparaginase II from E. coli K12 and Erwinia chrysanthemi is accompanied by side effects. It is desirable to search for other asparaginase I...
متن کاملEnhanced Expression of Recombinant Activin A in Escherichia coli by Optimization of Induction Parameters
Activin A is a member of the transforming growth factor β super family. Because of its extensive clinical usages, its recombinant production is beneficial. In this study, activin A was expressed in E. coli using the pET 21a expression vector. The optimization of the activin A production in E. coli was done by using the response surface methodology (RSM). At this stage, the effect of IPTG and la...
